CRISPR-Cas9 is an efficient, fast, easy technique for achieving knock-out gene in the cell. The mechanism has three steps: recognition, cleavage, and repair. The designed sgRNA directs Cas-9 and recognizes the target sequence. The Cas-9 nuclease is activated and makes double-stranded breaks (DSBs) when sgRNA recognizes the target sequence.
